چکیده

One method for recording neuronal activity is fluorescence microscopy, in which neurons in the brain express a fluorescent marker whose emission is modulated by the “firing” of the neuron. Our work is based on the integrated fluorescence microscope of the Schnitzer group [GBC11] which enables the acquisition of activity from large numbers of neurons in freely-behaving mice and rats. As shown in Fig. 1(a), the fluorescence microscope is surgically attached to the skull of a laboratory mouse. Fig. 1(b) shows a typical microscope image with individually outlined neurons. In a prototypical experiment, the animal is permitted to wander about its environment, while the microscope images the hippocampus – a region of the brain associated with spatial representation.

Imaging light responses of targeted neuron populations in the rodent retina.

Decoding the wiring diagram of the retina requires simultaneous observation of activity in identified neuron populations. Available recording methods are limited in their scope: electrodes can access only a small fraction of neurons at once, whereas synthetic fluorescent indicator dyes label tissue indiscriminately. Here, we describe a method for studying retinal circuitry at cellular and subce...

L- and D-cysteine functionalized CdS quantum dots as nanosensors for detection of L-morphine and D-methamphetamine

A new method in differentiation of chiral molecules is reported based on the fluorescence quenching of functionalized CdS quantum dots (CdS-QDs) as nanosensor by differing in the chirality of functionalization species. The chemically functionalized CdS-QDs with strong yellow emission were prepared using chiral L-cysteine (L-Cyst) and D-cysteine (D-Cyst) molecules. Then, the functionalized CdS-Q...

A Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...

Detection of abl/bcr Fusion Gene in Patients Affected by Chronic Myeloid Leukaemia by Dual-Colour Interphase Fluorescence in situ Hybridisation

Conventional cytogenetic is the standard technique for detection of Philadelphia (Ph) chromosome in chronic myeloid leukemia (CML). Evaluation of abelson murine leukemia/breakpoint cluster region (abl/bcr) fusion using dual-colour fluorescence in situ hybridization (D-FISH) is an alternative approach allowing rapid and reliable detection of the disease. We employed the technique of interphase D...

Encoding and decoding spatio-temporal information for super-resolution microscopy

The challenge of increasing the spatial resolution of an optical microscope beyond the diffraction limit can be reduced to a spectroscopy task by proper manipulation of the molecular states. The nanoscale spatial distribution of the molecules inside the detection volume of a scanning microscope can be encoded within the fluorescence dynamics and decoded by resolving the signal into its dynamics...
